O objetivo deste trabalho foi avaliar a influência da altitude e características químicas do solo sobre o perfil da comunidade de fungos micorrízicos arbusculares (FMA), presente em plantios de Coffea Arabica L. Amostras compostas de solo foram coletadas em uma profundidade de 0 a 10 cm em fazendas em Venda Nova do Imigrante - ES. A extração de DNA foi realizada com o kit Nucleo Spin Soil a partir de amostras de solo e posteriormente a técnica Nested PCR-DGGE foi utilizada para analisar o perfil da comunidade de FMA e também para o cálculo dos índices de diversidade. Amostras de solo também foram utilizadas para as análises químicas. As variáveis de equidade e diversidade diminuíram significativamente com a elevação da altitude. A saturação por bases (V (%)) influenciou positivamente a riqueza, equitabilidade e diversidade de FMA, enquanto a acidez potencial (H + Al) influenciou negativamente essas variáveis. A disponibilidade de Ca²⁺ resultou em aumento da riqueza de FMA. Altitude e fatores químicos do solo modulam a comunidade de fungos micorrízicos arbusculares do solo do cafeeiro. Assim, escolha do local de plantio e o manejo adequado do solo podem o aumento da diversidade destes fungos, que podem otimizar a absorção de água e sais minerais pelas raízes.
The objective of this study was to evaluate the influence of altitude and chemical characteristics of the soil in the profile of Arbuscular Mycorrhizal Fungi (AMF) community, present in Coffea Arabica L. plantations. Soil composite samples were collected at a depth of 0 to 10 cm in farms in Venda Nova do Imigrante - ES. DNA extraction were performed with the Nucleo Spin Soil kit from soil samples and subsequently the Nested PCR-DGGE technique. Equitability and diversity variables decreased significantly with elevation. Base saturation (V (%)) positively influences AMF richness, equitability and diversity, while potential acidity (H + Al) negatively influences these variables. The availability of Ca²⁺ resulted in increased MF richness. Altitude and soil chemical factors modulate the AMF community of coffee soil. Thus, the choosing of local of plantation and appropriate management can increase the diversity AMF, which an optimize mineral and water absorption by the plant roots. Coffee production has a great socioeconomic importance for Brazil. It generates direct and indirect jobs, and foreign exchange, with Brazilian Arabica coffee production estimated between 42 - 46 million bags (60 kg) in 2020. It is the main agribusiness activity in the State of Espírito Santo, Brazil with expected production between 13 - 15 million bags, and around 30% of this production is Arabica coffee. Technologies are recommended to coffee growers to increase yield, and production of specialty coffees on sustainable properties. Among the principles of integrated management is the monitoring of pests and diseases to determine the level of pest control. The estimate of the number of leaves to be sampled in the monitoring becomes an important tool to increase the accuracy of the obtained information. This research was carried out aiming to determine the minimum number of leaves necessary to evaluate the infestation of brown eye spot (BES) of coffee in Arabica coffee (Coffea arabica L.) without affecting the accuracy of the collection method. It was observed that the estimate of the minimum number for sampling was 46 leaves for the characteristics of incidence, and severity of BES in Arabica coffee. The modeling applied in this study allows to conclude that it is possible to recommend an optimum number of Arabica coffee leaves for these edaphoclimatic conditions, and variety, and it can serve as a basis for monitoring in an integrated pest and disease management program in Arabica coffee.
